Collagen is the most successful protein product that is not a protein product. It contains zero tryptophan, which means it is by definition an incomplete protein, and it is very low in leucine, which means it cannot drive muscle protein synthesis. Both facts are established, uncontroversial and entirely absent from the marketing. What collagen peptides can do is more specific and more interesting: hydrolysed collagen plus vitamin C, taken before loading, has repeatable data for tendon and ligament collagen synthesis (Shaw 2017; Baar's group), a modest but real signal for skin elasticity and hydration in meta-analysis (de Miranda 2021), and small joint-pain effects in athletes (Clark 2008; Zdzieblik 2017). It is a connective-tissue supplement. Sold as '20g of protein', it displaces protein your muscles could have used.

What the label is not telling you
- Collagen contains no tryptophan at all. That single fact makes it an incomplete protein by definition — it cannot be your protein source, and every gram counted toward a daily protein target as if it were whey is a gram of real protein you did not eat. This is the most consequential omission on the shelf and it is never printed.
- Leucine content is roughly a third of whey's. Collagen's amino profile is around a third glycine, plus proline and hydroxyproline — beautifully suited to building connective tissue matrix, useless for triggering muscle protein synthesis. Oikawa 2020 tested collagen peptides against whey in older women and found collagen failed to support muscle protein synthesis at matched doses.
- The tendon and ligament evidence is the strongest part of the category and depends on protocol, not brand. Shaw's 2017 work used ~15g hydrolysed collagen plus ~50mg vitamin C taken 60 minutes before loading exercise, with the loading being the actual stimulus. Take the same 15g on the sofa and you have taken an expensive glycine supplement. Nobody on the tub tells you the timing or the exercise requirement, because it makes the product conditional.
- Skin evidence is real and modest. The 2019–2021 meta-analyses find small improvements in elasticity and hydration at 2.5–10g/day over 8–12 weeks, with industry funding across most trials. Worth knowing; not worth the transformation photography.
- 'Multi-collagen from five sources' is a marketing structure, not a bioavailability upgrade. Your gut hydrolyses all of it into the same di- and tripeptides regardless of whether it started as bovine hide, chicken cartilage, fish scale or eggshell membrane. Type II from chicken cartilage has its own small cartilage literature at low doses; blending five sources at undisclosed ratios makes it impossible to know if any of them hit a studied dose.
- Glycine is arguably the most interesting ingredient and nobody markets it. A 15g collagen serving delivers roughly 3g of glycine, an inhibitory neurotransmitter with real sleep-onset and sleep-quality data at 3g (Yamadera 2007; Bannai 2012). If you feel better on collagen, glycine is the most likely explanation — and plain glycine costs a fraction of the price.

Effect on the nervous system
There are two real nervous-system stories here and neither is the one on the tub. The first is glycine: 3g of it is an inhibitory neurotransmitter at spinal and brainstem level, it lowers core body temperature, and lowering core temperature is one of the mechanical prerequisites for sleep onset. Yamadera's and Bannai's work showed 3g of glycine before bed improving subjective sleep quality and next-day cognition. A 15g collagen serving delivers roughly that. So the person who swears their evening collagen coffee calmed them down is probably right, and probably not for the advertised reason. The second story is the one that concerns us more: collagen displacing complete protein. Someone counting 20g of collagen toward a 100g daily target is functionally eating 80g, and the shortfall lands specifically on tryptophan — which is the only precursor for serotonin, most of it made in the gut and read by vagal afferents. Chase that for six months and the readout is a flatter mood, a worse-sleeping, shorter-fused version of yourself, arrived at while diligently tracking protein. This is the single most common protein-accounting error we see, and the category's labelling is why.
Who it might suit
Anyone rehabbing a tendon or ligament, taking 15g plus 50mg vitamin C 60 minutes before their loading exercise — that protocol has the best evidence in this review. Anyone with joint pain in a high-load sport at 10–15g/day for 12+ weeks. Anyone interested in the modest skin-elasticity effect at 2.5–10g/day. Anyone who wants the glycine sleep effect and does not mind paying a premium for it.
Who should skip it
Anyone counting it toward their protein target — that is the mistake this whole review exists to stop. Anyone expecting muscle gain. Anyone with a fish allergy taking marine collagen. Anyone taking it without the loading exercise and expecting tendon benefit. And anyone who could get the same glycine for a tenth of the price.
Bottom line
Collagen is a decent connective-tissue supplement and a bad protein. Use it if: you have a tendon or joint target — 15g plus 50mg vitamin C, 60 minutes before loading, for 12 weeks, from the cheapest tested brand. Do not: count a single gram of it toward your daily protein. Cheaper substitute for the effect most people actually feel: 3g of plain glycine before bed.
